Estradiol is a form of estrogen, a female sex hormone produced by the ovaries. Estrogen is necessary for many processes in the body.

Estradiol is used to treat symptoms of menopause such as hot flashes, and vaginal dryness, burning, and irritation. Other uses include prevention of osteoporosis in postmenopausal women, and replacement of estrogen in women with ovarian failure or other conditions that cause a lack of natural estrogen in the body. Estradiol is sometimes used as part of cancer treatment in women and men.

During and after the menopause, estrogens very efficiently remedy the consequences of estrogen deficiency, in particular the vasomotor symptoms ('hot flashes') and genitourinary atrophy. They also have a beneficial effect on incontinence and urinary tract infections. Estrogens also counteract an osteoporosis by reducing the calcium loss. However, fractures can only be avoided if a woman takes estrogens for a long period of time. Several epidemiological studies suggest that estrogens can advantageously influence cardiovascular morbidity and mortality, possibly through their effects on lipids. Estrogens have an established role in the hormone therapy of prostatic carcinoma.
